Thymosin Alpha-1

Thymosin Alpha-1 is a naturally occurring peptide originally isolated from thymic tissue. According to PubMed-indexed research, it acts through Toll-like receptors in dendritic cells and has demonstrated changes in immune cell subsets relevant to infection, cancer, and age-related immune decline (King & Tuthill, 2016). For patients dealing with chronic infections, post-viral syndromes, or immune dysregulation, it may be considered as part of a longevity protocol.

NAD+ (Nicotinamide Adenine Dinucleotide)

NAD+ is a coenzyme central to cellular energy production, DNA repair, and sirtuin activation, and levels decline with age. According to PubMed, "upregulation of NAD metabolism, including dietary supplementation with NAD precursors, has been shown to prevent the decline of NAD and exhibits beneficial effects against aging and aging-associated diseases" (Yaku et al., 2018). MOTS-c is a mitochondrial-derived peptide encoded by the 12S rRNA gene. Based on PubMed research, MOTS-c "prevented age-dependent and high-fat-diet-induced insulin resistance, as well as diet-induced obesity" in preclinical models (Lee et al., 2015). A 2023 review highlighted therapeutic potential for cardiovascular disease, metabolic syndrome, and age-related inflammation (Zheng et al., 2023). Human outcome data is limited, so this is best described as mechanistically interesting and investigational.

Epithalon

Epithalon is a synthetic tetrapeptide studied for its effect on telomerase. According to PubMed, Epithalon "prolonged the vital cycle of normal human cells due to overcoming the Hayflick limit" by inducing telomere elongation in aging fibroblasts in cell culture (Khavinson et al., 2004). Cell-culture telomere findings are a long way from a demonstrated longevity outcome in people, and any clinic presenting them otherwise is overstating the case.

Selank

Selank is a synthetic analog of the naturally occurring immunomodulatory peptide tuftsin. Based on PubMed-indexed research, Selank has demonstrated anxiolytic effects and modulates functional connectivity in brain regions associated with anxiety regulation, including the amygdala and temporal cortex (Panikratova et al., 2020).

Kisspeptin

Kisspeptin is a neuropeptide central to the hypothalamic-pituitary-gonadal axis, regulating release of gonadotropin-releasing hormone. It is used clinically to assess and support reproductive hormone function.

PT-141 (Bremelanotide)

PT-141 is a melanocortin receptor agonist that acts on the central nervous system rather than the vascular system. It is FDA-approved for hypoactive sexual desire disorder in premenopausal women, on the strength of two randomized phase 3 trials (Kingsberg et al., 2019; PMID 31599840).
